Sanger sequencing is the technique of choice if you need to determine the nucleotide sequence of individual DNA fragments, most often vector constructs or PCR amplicons. It is based on the separation of fluorescently labeled products of the sequencing reaction using capillary electrophoresis depending on their molecular weight.

We offer the following Sanger Sequencing protocols:

  • StandardSeq - This is the most common protocol in our Sanger sequencing portfolio of services. You are sending us a template that is ready for sequencing (purified).
  • RapidSeq - The fastest protocol in our portfolio of Sanger sequencing services. RapidSeq protocol samples are analyzed preferentially in the sequencers and their results are evaluated automatically.
  • ColonySeq - This is a protocol for direct plasmid sequencing from bacterial colonies and cultures, you send us an aliquot of culture or a bacterial colony picked from an agar plate as a template.
  • CleanSeq - This is a protocol for sequencing of PCR products, which also includes their enzymatic cleanup. You send us a template that is not ready for sequencing and must be purified first.
  • HairpinSeq - This is a special sequencing protocol for sequencing difficult templates. Bidirectional sequencing is included in this service. You send us a template that is ready for sequencing (purified).

Data analysis:

With the exception of the RapidSeq protocol, our specialists evaluate the results manually! Each result is checked before sending and re-analyzed as necessary to get the most out of it for you. We recommend that you pay attention to the articles in our Blog, where we continuously address various aspects of this method, including the evaluation of results.

StandardSeq

Standard single read sequencing service for plasmids or PCR fragments.

You supply:

  • PCR product or plasmid.
  • Template must be mixed together with the sequencing primer.
  • If you wish to use our universal primers, you supply the template only.

 

RapidSeq

Rapid sequencing service for plasmids or PCR fragments with automatic data analysis.

The fastest protocol in our portfolio of Sanger sequencing services. All samples delivered to our laboratory (for all protocols) are processed simultaneously, but samples for the RapidSeq protocol are analyzed preferentially in the sequencers and their results are evaluated automatically. You can expect first results in about 10 hours.

You supply:

  • PCR product or plasmid.
  • Template must be mixed together with the sequencing primer.
  • If you wish to use our universal primers, you supply the template only.

 

ColonySeq

Plasmid sequencing directly from bacterial colonies and cultures.

This is a protocol for direct plasmid sequencing from bacterial colonies and cultures, you send us an aliquot of culture or a bacterial colony picked from an agar plate as a template.

You supply:

  • The template is a bacterial suspension. In a separate tube you also supply sequencing primer(s). 
  • Only if you wish to use our universal primers, you supply the template only.
  • Please notice that count of primers you can use is limited (1 primer for every 8 samples).

 

CleanSeq

Sequencing of PCR products including their enzymatic cleanup.

This is a protocol for sequencing of PCR products, which also includes their enzymatic cleanup. You send us a template that is not ready for sequencing and must be purified first.

You supply:

  • The template is unpurified PCR product or other suitable template. In a separate tube you also supply sequencing primer(s). 
  • Only if you wish to use our universal primers, you supply the template only.
  • Please notice that count of primers you can use is limited (1 primer for every 8 samples).

 

HairpinSeq

Special sequencing protocol for hairpins, GC-rich and other problematic templates. The price includes sequencing from both directions.

This is a special sequencing protocol for sequencing difficult templates where the sequencing reaction can stop abruptly, typically due to the presence of some secondary structures and a sharp drop in signal may occur. Bidirectional sequencing is included in this service. You send us a template that is ready for sequencing (purified).
