Fragment analysis is a method of genetic analysis involving a series of techniques in which DNA fragments are fluorescently labeled, separated using capillary electrophoresis (CE), and their size is determined by comparison with a size standard.

Using our genetic analyzers, we offer fragment analysis for the following purposes, for example:

  • Authentication of cell lines
  • Microsatellite analysis (STR)
  • Determination of the efficiency of genome editing using the CRISPR-Cas9 technique
  • SNP genotypying and more

Fragment analysis makes it possible to find out information about the size of fragments, perform relative quantification or genotyping. The method is based on the preparation of fluorescently labeled DNA fragments by PCR using primers designed for a specific target region.

Typical graphic output of fragment analysis - size-separated color-coded peaks. On the y-axis, the fluorescence intensity is plotted for relative quantification, on the x-axis, the size of the DNA fragment in bp.

Benefits of using fragment analysis:

  • Multiplexing — alleles for overlapping loci can be distinguished using different fluorescently labeled specific primers. This makes it possible to analyze more than 20 loci in a single reaction.
  • Sensitivity - The method allows fragments differing by only one base pair to be separated.
  • Simple sample preparation
  • Easy data analysis
  • Fragment analysis does not require knowledge of the fragment sequence
  • Possibility of relative quantification

Capillary electrophoresis

Capillary electrophoresis of labelled DNA fragments, sizing typically up to 1200 bp. You supply PCR product or a mixture of PCR products (alternatively another suitable sample) which are fluorescently labelled.

We perform fragment analysis of DNA fragments labelled with different combinations of fluorescent dyes (dye sets). For selected dye sets we supply labelled size standard (see Sample requirements).

We perform manual data analysis. Results are typically available the same day we receive your samples.

Deliverables:

  • You can choose various combinations of up to five fluorescent dyes
  • Results online in .fsa format
  • Automatic email message as soon as your results are ready to download
  • Permanent internal quality control
  • Storage of your samples for one week, of your results for two years
  • Experienced technical support
